abstract The invention discloses a ratio type adhesive flare fluorescent probe and a preparation method thereof. The probe takes gold nanoparticles as a carrier, and DNA double strands with different fluorescent groups for identifying telomerase are connected to the surface of the probe: FL-R flare long chain and SH-C-FL strand short chain with a sequence complementary to the long chain part. The FL-R flare long chain is sequentially provided with a fluorescent dye, an auxiliary sequence, a sequence complementary with a telomerase amplification sequence and a telomerase primer sequence from the 5' end, and the FL-R flare long chain emits light no matter whether telomerase exists or not and can be used as an internal fluorescent signal reference. One end of the SH-C-FL strand is modified with sulfydryl, the other end of the SH-C-FL strand is modified with another fluorescent dye, in the presence of telomerase, the short chain is extended to form a hairpin structure, the hairpin structure falls off from nanogold, fluorescence is turned on, the hairpin structure is used for carrying out in-situ detection on telomerase activity in cells, and the quantitative analysis on the telomerase activity in tumor cells and the effective distinguishing of the tumor cells from normal cells can be realized by combining long-chain fluorescent signals. The probe does not need to be cleaned in the cell experiment process, and can be directly used for laser confocal imaging.
