abstract | The invention relates to a method for preparing 10-hydroxy-2-decenoic acid by using resting cells of escherichia coli engineering bacteria, which comprises the following steps: (1) constructing an escherichia coli engineering bacterium containing a recombinant plasmid pET-28 a-ydiI; (2) preparing an induced cell from an escherichia coli engineering bacterium containing a recombinant plasmid pET-28 a-ydiI; (3) culturing the induced cells in a transformation culture medium to obtain resting cells, and then adding 10-hydroxydecanoic acid into the culture medium to prepare 10-hydroxy-2-decenoic acid; the invention discloses positive recombinant escherichia coli containing thioesterase, which is modified by genetic engineering for the first time, and can produce 10-hydroxy-2-decenoic acid through resting cell fermentation after special induction treatment, thereby realizing the mass biosynthesis of 10-hydroxy-2-decenoic acid. |
