titleOfInvention | Identification-enhanced metastable aptamer probe and application thereof in counting analysis of target substances |
abstract | The enhanced metastable state nucleic acid aptamer recognition probe has the melting temperature Tm lower than 45 ℃ and is a hairpin structure with complementary tail ends; when the target substance exists, the nucleic acid aptamer probe can be combined with the target substance, the tail end stem part of the nucleic acid aptamer probe is further opened, the rolling circle amplification reaction is triggered through the action of polymerase, when the target substance does not exist, the nucleic acid aptamer probe forms a closed structure, the double-stranded structure of the nucleic acid aptamer probe is more stable through extension of the tail end stem part, and the rolling circle amplification reaction cannot be triggered under the action of the polymerase. The preferred recognition enhancement type metastable state aptamer probe has a nucleotide sequence shown as SEQ ID NO: 1. SEQ ID NO: 2 or SEQ ID NO: 3, the preparation method is as follows. The recognition enhancement type metastable state aptamer probe is applied to counting analysis of target substances, response sensitivity and recognition efficiency of target molecules can be improved, and the recovery rate reaches 88-110%. |
