abstract | The invention relates to a preparation method of dronedarone hydrochloride. The method comprises the steps of using 1, 4-cyclohexanedione as a raw material, carrying out a halogenation reaction on the raw material and chlorine or bromine to generate dihalogen-1, 4-cyclohexanedione, carrying out a nucleophilic substitution reaction, an elimination reaction and a dehydration cyclization reaction on the dihalogen-1, 4-cyclohexanedione and 1- [4- (3-di-n-butylamino-propoxy) -phenyl ] -heptane-1, 3-dione to generate a compound V and a compound V', ammoniating the compounds with ammonia gas, carrying out methylsulfonyl chloride sulfonylation on the obtained product to generate dronedarone hydrochloride, and salifying the dronedarone hydrochloride with hydrochloric acid to generate dronedarone hydrochloride. The preparation method disclosed by the invention is simple in process flow, few in operation steps, low in production cost, high in yield and purity, small in waste water generation amount, green and environment-friendly, and beneficial to industrial production and application of dronedarone hydrochloride. |
