abstract The invention discloses an extraction method and application suitable for obtaining a large amount of high-purity salmonella outer membrane protein. The method comprises the following steps: (1) cloning the gene of the outer membrane protein of the salmonella into an expression vector, and then introducing the obtained recombinant expression vector into host salmonella to obtain a recombinant strain; (2) carrying out amplification culture on the recombinant strain, then centrifugally collecting thalli, and freezing and storing to obtain frozen thalli; (3) adding a protein lysate into the frozen bacteria, adding a phenylmethylsulfonyl fluoride solution, carrying out constant-temperature ultrasonic crushing, and collecting protein precipitate; (4) adding outer membrane protein extracting solution into the protein precipitate, uniformly mixing, incubating at 4 ℃, centrifuging, and collecting supernatant; (5) separating and purifying the supernatant to obtain the salmonella outer membrane protein. The method has simple steps and convenient operation, is suitable for extracting a large amount of high-purity and high-activity outer membrane protein, and can simultaneously meet the requirements of subsequent experiments on protein activity, structure, vaccine research and development and the like.
