The present invention obtains a maltogenic amylase mutant with improved transformation efficiency through site-directed mutation and combined mutation, and expresses the mutant in Escherichia coli. The enzyme liquid was used for maltose production. Under the addition of 20U/g dry starch, the mutant K198E/D290T in the secondary saccharification process could increase the maltose content to more than 97% when the conversion reaction was carried out for 10h, trisaccharide, tetrasaccharide. The content approached 0 when the reaction was 5h. Compared with wild-type maltogenic amylase, it has higher conversion efficiency and fewer conversion by-products, which has important industrial application potential. |
