abstract The invention provides a method for determining monosaccharide composition in acidic polysaccharide, which comprises the following steps: (1) performing first-step hydrolysis on polysaccharide through weak acid, and then performing second-step hydrolysis by matching with strong acid; (2) monosaccharide composition in the hydrolyzed sample was determined by ion chromatography in combination with an electrochemical detector. The weak acid in the first hydrolysis step is trifluoroacetic acid with lower molar concentration, and the strong acid in the second hydrolysis step is sulfuric acid with higher molar concentration. According to the obtained ion chromatogram, the type and content of monosaccharide can be obtained based on peak migration time and peak area. Compared with the traditional one-step hydrolysis method, the method has high accuracy and good selectivity, and is suitable for analyzing the monosaccharide composition of the acidic polysaccharide.
