abstract | Methods and compositions are disclosed for preparing DNA libraries from slide-mounted FFPE tissue samples for downstream next-generation sequencing. In one embodiment, a method of preparing a DNA library is disclosed. The method may include applying droplets of the reagent mixture to a slide-mounted FFPE tissue sample. The reagent mixture may comprise one or more buffers, cofactors, non-ionic surfactants, glycerol solution, gelatin solution, dNTPs, DNA polymerase, and a primer pool comprising a plurality of forward and reverse primers. The method may also include stirring the droplets in a circular motion on the glass slide while scraping the portion of the FFPE tissue sample mounted on the glass slide to generate a reaction mixture. The method may further comprise aspirating the reaction mixture from the slide directly into a pipette tip and dispensing the reaction mixture into reaction vessels for further amplification. |
