The present invention relates to a genetic engineering bacterium producing heparosan. The genetically engineered bacteria use food-grade Corynebacterium glutamicum as the host cell. By introducing the heparosan synthase gene, co-expressing the key genes of the heparosan synthesis pathway, and knocking out or weakening the genes related to the competitive metabolic pathway, the genetic engineering of the Corynebacterium glutamicum is carried out. It is obtained by transforming microorganisms, introducing lyase genes, and regulating the enzymatic activity of heparosan synthase in Corynebacterium glutamicum or heparosan-producing genetically engineered bacteria by means of RBS regulation strategy or enzyme modification. The engineered bacteria are safe, high-yield, non-toxic, and capable of synthesizing low-molecular-weight heparosan. |
