abstract | The invention belongs to the technical field of plant genetic engineering, in particular to a cotton wilt resistance gene and its application. From the identification of fusarium wilt resistance in cotton population and genome-wide association analysis, the upland cotton fusarium wilt resistance gene was located in the 400kb genome interval of chromosome 17, 1.7-2.1Mb. Virus-mediated gene silencing technology was used to inhibit the expression of all genes in this genomic region, and after inoculation and identification, the major cotton fusarium wilt resistance gene GhFov1 was screened. The gene, encoding a glutamate receptor protein kinase, was silenced in resistant upland cotton and significantly reduced resistance to Fusarium wilt. Transforming tobacco found that GhFov1 protein could recognize cotton wilt and activate immune response to produce allergic necrosis. A pair of primers that can detect GhFov1 genotype in Upland cotton was developed. The primers and the optimized PCR reaction system can be used to evaluate the resistance to fusarium wilt of cotton germplasm resources and varieties. |
