abstract | The invention discloses a preparation method of an oral insulin preparation taking medicinal mulberry leaf polysaccharide as a carrier, which comprises the following steps of (1) constructing a medicinal mulberry leaf polysaccharide-phenylboronic acid carrier; (2) self-assembly of the medicinal mulberry leaf polysaccharide-phenylboronic acid carrier nano-carrier; (3) preparation of oral insulin preparation. The mulberry leaf polysaccharide is used as a carrier material, phenylboronic acid with glucose sensitivity is used for modifying the mulberry leaf polysaccharide, the mulberry leaf polysaccharide can be self-assembled into a nano carrier and then can be used for loading and conveying insulin serving as a hypoglycemic drug, and the polysaccharide carrier and the insulin can play a synergistic hypoglycemic role after oral administration. The insulin can be slowly released under the protection of the mulberry leaf polysaccharide-phenylboronic acid nano-carrier, the gastrointestinal acidic condition has little influence on the mulberry leaf polysaccharide-phenylboronic acid nano-carrier, the accumulated release amount of the insulin in 12 hours reaches 60%, and the insulin-releasing agent has responsiveness to glucose, can release the insulin under the high-sugar condition and reduces the blood sugar. |
