abstract The invention discloses a liquid-mass spectrometry method for measuring acetyl-CoA in the liver of an animal, in particular, first taking fresh liver of the animal, rinsing the blood in the liver, and then mixing the liver sample with the pre-treatment liquid, and then homogenizing. Tissue homogenate, add internal standard in the homogenate, mix evenly and centrifuge, take the supernatant as the sample to be tested, and finally send the sample to the liquid chromatography-mass spectrometer for detection, collect data, and return to the standard curve. The concentration of acetyl-CoA in the liver of the animal is calculated, wherein the pre-treatment liquid is an aqueous perchloric acid solution containing dithiothreitol and phenylmethylsulfonyl fluoride. Compared with the existing enzyme-linked immunolabeling method, the liquid-mass method used in the invention is fast, reliable, and has simple operation steps, and is suitable for batch determination of the content of acetyl-CoA in the liver.
