The invention discloses a construction and application of a recombinant strain for producing L‐2-aminobutyric acid by converting L-threonine, and belongs to the technical field of bioengineering. The production method of the present invention utilizes a recombinant plasmid expressing a double plasmid, and simultaneously realizes high-efficiency expression of three enzymes, converts L-threonine into L-2-2 aminobutyric acid, and couples a coenzyme regeneration system. By converting NAD+ to NADH, the concentration of NADH in the system is relatively stable, and the transformation can be carried out efficiently. Further, the CO2 converted from ammonium formate in the process is soluble in the ammonia water formed in the conversion system, has little environmental pollution, and has industrial application value. The method has the advantages of mild transformation conditions, strong specificity, low cost and short conversion time. The present method was used to prepare L-2-aminobutyric acid, and 40 g/LL-threonine was added. The concentration of the product L-2-2-aminobutyric acid was 43.3 g/L, and the conversion rate was over 99.9%. |
