titleOfInvention Polycyclic aromatic hydrocarbon degrading engineering bacterium and engineering modification method and application thereof
abstract The invention relates to a polycyclic aromatic hydrocarbon degrading engineering bacterium and an engineering modification method and application thereof. The gene catA2 which is a degradation gene for coding the cyclolytic dioxygenase is obtained by cloning pseudomonas aeruginosa SA1, and the nucleotide sequence is shown as SEQ ID No. 1; the nucleotide sequence of the specific oprL promoter is shown as SEQ ID No. 2; selecting a broad-host plasmid pBBR1MCS-5 as a vector; the catA2 gene and the oprL promoter are respectively cloned into a pBBR1MCS-5 vector, transferred into a receptor cell for cloning and amplification, and positive clones are screened to obtain a target recombinant vector pBBR1MCS-5-oprL-catA 2; the pseudomonas aeruginosa SA1 is used as a host, and the recombinant vector pBBR1MCS-5-oprL-catA2 is transformed into pseudomonas aeruginosa strains by an electric shock method to construct engineering pseudomonas aeruginosa. Has the capability of degrading polycyclic aromatic hydrocarbon.
