abstract The present invention is based on the surprising discovery that linkers used in bioconjugates, such as antibody-drug conjugates, are not therapeutically inert, but have an impact on the therapeutic index of the bioconjugate. Accordingly, the present invention relates to methods of increasing the therapeutic index of bioconjugates and bioconjugates for use in the treatment of, in particular, cancer. Bioconjugates according to the present invention have a sulfonamide linker comprising a group according to formula (1).
