abstract |
The invention relates to a novel preparation method of nucleoside modified 3'-O-CH 2 N 3 -2'-O-Me-cytidine nucleoside. The method of the present invention uses the natural nucleoside modified 2'-O-Me-N 4 -Bz-cytidine nucleoside as a raw material, first reacts with a silylating agent, and silylates the 5'-OH; Anhydride reaction yields intermediate 3'‑O‑CH 2 SCH 3 ‑2'‑O‑Me‑N 4 ‑Bz‑cytidine; subsequent treatment with sulfuryl chloride yields key intermediate 3'‑O‑CH 2 Cl ‑2'‑O‑Me‑N 4 ‑Bz‑cytidine nucleoside; then through azide substitution, deprotection and other processes, high yield and high selectivity to generate the desired new nucleoside modification 3'‑ O ‑ CH2N3‑2' ‑O‑Me‑cytidine. The method of the invention is simple and economical and can realize large-scale production. |
