abstract The invention relates to a method and a kit for in vitro detection of MUSK-Ab and LRP 4-Ab. The method comprises the following steps: taking 293T cells, and transfecting the 293T cells with a pCMV6-AC-GFP-MuSK recombinant plasmid and/or a pCMV6-AC-GFP-LRP4 recombinant plasmid; adding paraformaldehyde into transfected cells transfected with the pCMV6-AC-GFP-MuSK recombinant plasmid and/or transfected cells transfected with the pCMV6-AC-GFP-LRP4 recombinant plasmid, and fixing for 15-30 min; after washing with phosphate buffer solution, adding bovine serum albumin and TritonX-100 for sealing and permeating for 2-3 h; adding the serum to be detected into the transfection cells of the closed and permeable pCMV6-AC-GFP-MuSK recombinant plasmid and/or the transfection cells of the closed and permeable pCMV6-AC-GFP-LRP4 recombinant plasmid, incubating for 45min to 2h at 24 to 28 ℃, washing by phosphate buffer solution, and adding Alexa Fluor TM 568 labeled goat anti-human IgG is incubated at 24-28 ℃ for 45 min-2 h, washed by phosphate buffer solution, excited by blue excitation light and green excitation light of a fluorescence microscope, observed, photographed and recorded. The used reagents are all conventional reagents, and the detection cost is low without depending on expensive kits.
