abstract The invention relates to a fluorescence labeling cleavable nucleotide which is characterized by having a structure shown in the following general formula (I); in the general formula (I): fluororescent represents a fluorophore; base represents a Base; r 1 ~R 8 Each independently selected from hydrogen atom, C 1 ~C 10 Alkyl of (C) 1 ~C 10 An alkoxy group of (a), a PEG chain, a carboxylate, a substituted or unsubstituted phenyl; m is an integer of 0 to 10; n is an integer of 0 to 10. The invention also provides a preparation method of the fluorescence labeling cleavable nucleotide. The cleavable nucleotide based on the disulfide bond linkage unit provided by the invention has great potential and value in DNA sequencing; meanwhile, the synthesis method of the cleavable nucleotide has the characteristics of simple and convenient operation, low requirement on equipment, low cost, high equipment use efficiency and the like, and meets the requirement of large-scale production and application.
