abstract The present invention relates to a novel preparation method of nucleoside modifier 5'-DMTr-2'-EOE-thymidine. The method of the present invention uses the boronic acid ester modified by the specific target group as the source of the alkylated group of the sugar ring hydroxyl group, and uses cesium carbonate as a catalyst to react with the dehydrothymidine protected by 5'-DMTr-protected, high Yield, highly selective generation of target product 5'-DMTr-2'-EOE-thymidine. The method of the invention can easily and economically synthesize 5'-DMTr-2'-EOE-thymidine in large quantities.
