abstract | The invention provides a polycation gene vector, a preparation method and application thereof, wherein the polycation gene vector comprises a grafting framework and arginine protected by tosyl grafted with the grafting framework; the raw material of the grafting framework is selected from dendritic polyamide-amine, hyperbranched polylysine with molecular weight of 2000-100000 g/mol or linear epsilon-polylysine with molecular weight of 2000-50000 g/mol; the dendritic polyamidoamine is selected from one or more of model PAMAM-G3, model PAMAM-G4 and model PAMAM-G5. The polycation gene vector provided by the invention grafts the arginine protected by the tosyl on a specific grafting framework, and can obviously improve the transfection efficiency of the compound nanoparticles prepared by the polycation vector. In addition, the charge density of the nano-composite particles can be effectively reduced, and the cytotoxicity of the nano-composite particles is further reduced. |
