abstract | The invention discloses a polypeptide of a targeting Syntenin protein, a dimer thereof and application thereof, belonging to the technical field of biological medicine. The amino acid sequence of the polypeptide is Cys-Asp-Lys-Glu-phi-Val, wherein phi is 3- (2-naphthyl) -L-alanine. The polypeptide provided by the invention is obtained by optimizing amino acid of screened natural polypeptide ligand, and the affinity of the polypeptide and a Syntenin protein tandem PDZ structural domain is obviously improved. The dimeric polypeptide obtained by dimerizing the polypeptide has the affinity with a tandem PDZ structural domain as high as 210nM, can effectively inhibit migration, proliferation and invasion of various cancer cells with high Syntenin expression under the concentration of 10uM, does not generate other toxic or side effects, and provides a design thought for drug development for treating the malignant tumor metastasis and invasion with high Syntenin expression. |
