abstract |
This article provides a solution to the problem of heavy and light chain mismatches in bispecific antibodies. Part of the solution involves the Fc region engineered in its CH3 domain by lysine repositioning to drive the heterodimerization of the two heavy chains of the bispecific antibody. The second part of the solution involves the modification of one of the two Fab arms of the bispecific antibody in order to prevent mismatching of the light chains in the bispecific antibody. In particular, the CH1 and CL domains of one of the Fab arms of the bispecific antibody are replaced by an IgE CH2 domain or an IgM CH2 domain. |
