abstract | Sildenafil has a good effect on treating male erectile dysfunction (ED) clinically. There are many reports on the preparation method of sildenafil. Most of them are two-step synthesis, and the two-step post-processing operation is relatively cumbersome, and the yield lower. The invention provides a method for synthesizing a sildenafil intermediate. Compound II is dissolved in an organic aprotic solvent, and compound I is added for condensation reaction; after the reaction of compound II is completed, a base is added for cyclization reaction to obtain compound IV. The invention adopts the "one-pot method" synthesis, reduces the generation of "three wastes", greatly shortens the reaction time, improves the reaction yield, and has good stability. |
