abstract The invention discloses a method for screening miR-3880 target genes, which takes goat genome DNA as a template and carries out the steps of Taq DNA polymerase, buffer environment and Mg 2+ In the presence of dNTPs, performing amplification by using a primer OGR1 under the PCR condition, and determining the obtained PCR product as a sequence of an OGR1 gene 3' UTR region; then constructing a dual-luciferase report system, detecting luciferase activity, and preliminarily identifying a target gene of miR-3880; detecting the influence of the miR-3880 analogue on the mRNA level of the OGR1 gene by adopting an RT-qPCR method; and detecting the influence of the miR-3880 analogue on the protein level of the OGR1 gene by using a Western blot method. The binding site of the OGR 13' UTR region and miR-3880 is determined, the targeting regulation and control relation between miR-3880 and a target gene OGR1 is verified, miR-3880 can inhibit the expression of the OGR1 gene on the mRNA and protein level, and the OGR1 is further confirmed to be the target gene of miR-3880. Lays a foundation for researching the influence of miR-3880 on the mammary gland development and lactation function of the dairy goat.
