abstract The invention discloses a recombinant TM4 phage library for constructing a glutamic acid gene family deletion mycobacterium tuberculosis and application thereof. The invention firstly provides a recombinant shuttle vector library required for constructing the recombinant TM4 phage library, wherein the recombinant shuttle vector library comprises 19 recombinant shuttle vectors which are respectively inserted into homologous arm sequences at two sides of 19 member genes of a mycobacterium tuberculosis glutamic acid family. The recombinant TM4 phage library constructed by the invention can be used for knocking out the genes, verifying whether the genes are necessary genes of the mycobacterium tuberculosis, constructing gene mutation replacement strains, constructing gene knock-out mutant strains, researching the enzymatic activity function of the mutant genes after clinically separating the gene mutation, and providing potential targets for the development of amino acid metabolism drugs of the mycobacterium tuberculosis.
