abstract | The invention discloses an application of clematis root in the separation preparation of 5-hydroxymethylfurfural, which comprises the following steps: extracting radix Clematidis with acidic solution, concentrating, centrifuging, and lyophilizing to obtain crude extract; dissolving the crude extract in methanol solution, centrifuging, collecting supernatant, concentrating, and dissolving in water to obtain crude extract containing 5-hydroxymethylfurfural; adsorbing the crude extract with macroporous resin, eluting, detecting with high performance liquid chromatography, mixing eluates, and concentrating to obtain concentrated extract; preparing the enrichment by adopting a semi-preparative high performance liquid chromatography, collecting fractions containing 5-hydroxymethylfurfural, concentrating and freeze-drying to obtain the 5-hydroxymethylfurfural. The invention extracts 5-hydroxymethyl furfural in clematis root by acid solution, and adopts a macroporous resin separation method combined with a high performance liquid chromatography and a semi-preparative high performance liquid chromatography to realize the separation and preparation of 5-HMF from clematis root. |
