abstract The invention discloses a method for constructing a spontaneous anemia mouse model with leukocytosis and thrombocytosis, which carries out site-specific mutagenesis on the 177 th lysine and the 180 th lysine of mouse STAT3 protein to arginine. According to the invention, point-directed mutation is carried out on STAT3 gene, and 177 th lysine and 180 th lysine of STAT3 are subjected to point-directed mutation to arginine, so that the mice are found to be an anemia model in which white blood cells including lymphocytes, granulocytes and monocytes are increased, platelets are increased, but red blood cells and hemoglobin are obviously reduced. The disease mouse model established in the invention has important significance for researching the generation and development mechanism of anemia accompanied with the rise of leucocytes and blood platelets and developing antagonistic drugs.
