abstract The invention provides a culture method of gamma T cells, which comprises the following steps: (1) obtaining mononuclear cells from peripheral blood, carrying out heavy suspension by using serum-free culture medium additionally added with antibody, adjusting the density of the mononuclear cells, and culturing in an incubator; (2) on day 4 and day 8, serum-free culture media without the addition of antibodies were supplemented respectively; on day 12, the amplification culture is finished; (3) screening the T cells, transferring the sorted gamma T cells to a cell culture bottle, and supplementing a gamma T cell culture medium; (4) on day 4 and day 8, the gamma T cell culture media were supplemented, respectively; on day 12, the amplification culture is finished; (5) the gamma T cells were collected. The invention has the advantages that: provides a cell culture method specially aiming at gamma T cells, the gamma T cells are amplified for 12 days, the amplification multiple reaches 60 times, and the killing activity of the cultured gamma T cells on K562 tumor cells reaches more than 90 percent when the ratio is 25: 1.
