abstract | The invention discloses a synthesis method of 4-bromo-D-phenylalanine, which comprises the following steps: 1) adding diethyl acetamidomalonate, sodium ethoxide and ethanol into a reaction bottle, raising the temperature to 80°C, and keeping the temperature React for 2 hours, cool down to 35°C and add p-bromobenzyl bromide, keep the temperature for 5h, cool down to 25°C, concentrate and crystallize under reduced pressure to obtain diethyl (acetamido)[(4-bromophenyl)methyl]malonate, Add sodium hydroxide to it, raise the temperature to 45°C and keep it warm for 4 hours, cool down to 20°C to crystallize (acetamido)[(4-bromophenyl)methyl]malonic acid; add 60%W/V Heat up the hydrochloric acid solution to 85°C and keep it warm for 2 hours. After the reaction is completed, the concentrated acid crystallizes to obtain 4-bromo-D-phenylalanine. The invention has relatively mild reaction temperature and safe operation, and can obtain products with higher purity compared with traditional techniques, and the raw materials used are relatively cheap and easy-to-obtain raw materials, and the cost is also very low. |
