abstract | The invention discloses L-rhamnose gum-1-phosphate aldolase and application thereof in catalytic synthesis of rare sugar D-psicose, wherein the gene sequence of the L-rhamnose gum-1-phosphate aldolase is that the wild L-rhamnose gum-1-phosphate aldolase coded by SEQ ID NO.1 is subjected to saturation site-directed mutagenesis at the 29 th, 188 th, 266 th or 152 th positions of amino acid of the L-rhamnose gum-1-phosphate aldolase respectively. The invention overcomes the defects that D-psicose is generated by wild RhaD, wherein D-sorbose is 1: 1 has no defect of stereoselectivity, so that the catalytic reaction only produces D-psicose or mainly produces D-psicose, and the enzyme activity of the mutant RhaD is higher. The method for preparing D-psicose from the RhaD with site-specific mutagenesis, which is prepared by the method, has the advantages of high synthesis efficiency, simple process, cost saving and highest D-psicose catalysis efficiency of 100%, and realizes the large-scale synthesis of D-psicose from D-glyceraldehyde under the catalysis of the RhaD in the industrialization. |
