titleOfInvention | Cas9 nuclease-based DNA detection and analysis method and application thereof |
abstract | The invention provides a DNA detection and analysis method based on Cas9 nuclease and an application thereof, and the method comprises three steps: (1) performing PCR amplification on the target DNA; (2) treating PCR amplification by CAT method; (3) PCR amplification was performed using CAT-treated DNA as a template. The method can be used for successfully detecting the L1 and E6/E7 genes of HPV16 and HPV18 in human cervical carcinoma cells. The invention utilizes the specificity recognition cutting characteristic of the CRISPR technology to DNA, and successfully avoids the key bottleneck problems of nucleic acid hybridization, specificity PCR primer design and the like in the field of nucleic acid detection and typing at present. |
