abstract | The invention provides application of E3 ligase FBW7 in preparation of a medicine for inhibiting telomere damage, delaying senescence and preventing and treating senescence-related diseases, wherein the amino acid sequence of FBW7 is shown as SEQID No. 2. Research proves that gene expression of FBW7 is inhibited by knockout or RNA interference, or FBW7 gene mutant is expressed to competitively inhibit FBW7 protein, or TPP1 is over-expressed to overcome degradation caused by FBW7, so that stress such as radiation, chromosome telomere damage caused by bleomycin and hydrogen peroxide can be protected, TPP1 is prevented from being degraded in a basic state or a stress state, the effect of lengthening the length of telomere is caused, the number of tissue stem cells is increased, the lung respiratory function in the basic state is improved, lung tissue senescence and pulmonary fibrosis in the stress state are prevented, and the application of the composition in preparing a medicament for preventing and treating telomere dysfunction can be realized. The invention provides a new idea and means for resisting aging. |
