abstract | A micro-fluidic chip-based metabolism-dependent drug cardiotoxicity evaluation method uses a visual transwell micro-fluidic chip, wherein the upper layer is inoculated with hepg2 cells, and the lower layer is inoculated with human induced pluripotent stem cells, and simultaneously carries out in-situ induced differentiation on cardiomyocytes; the material of the micro-fluidic chip is a light-permeable and air-permeable polydimethylsiloxane polymer, the ratio of polydimethylsiloxane monomers to an initiator is (15-5): 1, the material of the porous filter membrane is a polycarbonate membrane, and the aperture of the polycarbonate membrane is 0.01-10 um; the lower surface of the top chip of the microfluidic chip is irreversibly sealed with the porous filter membrane, and the upper surface of the bottom chip is bonded with the porous filter membrane by polydimethylsiloxane. The invention uses the micro-fluidic chip as a platform, firstly constructs a model for evaluating the myocardial toxicity of the metabolism-dependent drug close to physiological conditions in vitro, and provides an important platform for developing and screening the metabolism-dependent drug. |
