abstract | The invention discloses an improved method for preparing lamotrigine, which comprises the following steps: step I: 3-amino-1, 2, 4-triazine (I) and a halogenating reagent are subjected to halogenation reaction in the presence of an organic solvent to obtain 3-amino-6-halogeno-1, 2, 4-triazine (II); step II: the compound II is subjected to amination reaction to prepare 3, 5-diamino 6-halogeno-1, 2, 4-triazine (compound III); step III: the compound III and 2, 3-dichlorobenzene boric acid derivative are subjected to suzuki coupling reaction in a solvent under the catalysis of palladium/[ N, N ] pyridine amidino ligand/alkali to prepare the lamotrigine (IV). The method does not use metal cyanide, and has better reaction selectivity and higher reaction yield. |
