titleOfInvention Preparation method of rabbit anti-polyclonal antibody for specifically recognizing jujube witches broom phytoplasma and application kit thereof
abstract The invention belongs to the field of jujube witches broom phytoplasma detection, and particularly relates to a preparation method of a rabbit anti-polyclonal antibody for specifically recognizing jujube witches broom phytoplasma and an application kit thereof. The rabbit anti-polyclonal antibody for specifically recognizing jujube witches broom phytoplasma provided by the invention is mainly applied to the detection of jujube witches broom, and the preparation method comprises the following effective steps: a. cloning from total DNA of jujube tree infected with jujube witches broom phytoplasma to obtain an immune membrane protein gene sequence of the jujube witches broom phytoplasma, predicting an amino acid sequence of the immune membrane protein, selecting a 47-152aa region with good C-terminal epitope exposure for protein expression, and performing label purification; b. after the tag is purified, the selected protein is mixed with incomplete Freund's adjuvant to immunize a white rabbit, the blood of the rabbit is collected, and the polyclonal antibody of the jujube witches broom phytoplasma immune membrane protein is obtained through antigen affinity purification. The rabbit anti-polyclonal antibody for specifically recognizing the jujube witches broom phytoplasma provided by the invention has obvious specificity in the detection of the jujube witches broom, and the detection accuracy is ensured.
