abstract | The invention discloses a method for establishing an osteoarthritis cell model after directional differentiation of mesenchymal stem cell cartilage by a two-step method. The method firstly induces mesenchymal stem cells to directionally differentiate into articular chondrocytes, and then adopts IL-1 beta to induce the obtained chondrocytes in vitro to establish an osteoarthritis model. The invention firstly uses a two-step method to establish an osteoarthritis model after cartilage directional differentiation, and determines the optimal use concentration of a solution and the cell induction time by inspecting the morphology of chondrocytes and the expression change of phenotypic genes aggrecan and Col2 alpha 1. The method is easy to operate and low in cost, and can be used for cell quality evaluation after directional differentiation of the mesenchymal stem cell cartilage, early warning of adult osteoarthritis susceptibility, verification of osteoarthritis drug action targets, drug toxicity/drug effect screening and other related researches. |
