abstract The invention discloses a kind of methods that two-step method establishes osteoarthritic cells model after mescenchymal stem cell cartilage directed differentiation.This method inducing mesenchymal stem cell directed differentiation first is articular chondrocytes, then establishes osteoarthritis using the external evoked gained cartilage cells of 1 β of IL.The present invention establishes osteoarthritis after cartilage directed differentiation using two-step method for the first time, is changed by investigating cartilage cell's morphology and phenotype genes aggrecan, Col2 α 1 expression, it is determined that the optimal use concentration of solution and cell induction time.This method operation easily, cost it is relatively low, available for correlative studys such as cell quality assessment, the susceptible early warning of adult osteoarthritis, the verification of osteoarthritis drugs action target and the screenings of drug toxicity/drug effect after mescenchymal stem cell cartilage directed differentiation.
