abstract The invention belongs to the technical field of environmental pollution detection, and relates to a design method and application of a phthalate ester environmental hormone universal structure hapten, which comprises the following steps: (1) comprehensively utilizing molecular simulation software such as Avogadro and Gaussian 09 and the like to simulate the geometrical structures, charge distribution and molecular hydrophobic surface conditions of 6 phthalate ester environmental hormones and 5 hapten molecules, comprehensively evaluating the results, and screening out the optimal immune hapten (hapten 1) and the coating hapten (hapten 5); (2) coupling hapten1 and bovine serum albumin by a diazotization method to synthesize immunogen; (3) coupling hapten5 and ovalbumin by using an active ester method to synthesize a coating antigen; (4) preparing polyclonal antibody and measuring antibody characteristics. The invention successfully designs the phthalate general hapten by using a molecular simulation technology, and verifies through an animal immunity experiment, thereby proving that the method can greatly improve the research and development efficiency of the hapten, save time and money consumption, have good prediction capability and provide reference for the design of other micromolecular haptens.
