titleOfInvention Purification and concentration method and system for recombinant avian influenza virus
abstract The invention relates to the field of virus purification and concentration, in particular to a purification and concentration method and a purification and concentration system for recombinant avian influenza virus. A purification and concentration method of recombinant avian influenza virus comprises the following steps: centrifuging the pretreated recombinant avian influenza virus to remove large granular substances and cell debris to obtain clear liquid; treating the clear liquid by using a hollow fiber microfiltration system to obtain permeate; treating the permeate with membrane-packed ultrafiltration concentration system, collecting the circulating solution, and concentrating. The purification and concentration method provided by the invention comprises the steps of firstly removing cell debris and large-particle substances through high-speed centrifugation, purifying and clarifying through a hollow fiber microfiltration system to remove small-particle proteins, and then concentrating through a membrane-packed ultrafiltration concentration system to obtain the purified and concentrated recombinant avian influenza virus. The whole process is simple and easy to implement, the impurity content and the impurity protein content in the virus are effectively reduced, and the impurity protein content in the virus is removed by more than 90%; after concentration, the virus titer is obviously improved.
