abstract A liver-blood brain barrier system for simulating in vivo metabolic process based on a microfluidic chip mainly comprises a top chip, a porous filter membrane and a bottom chip. The top chip is inoculated with liver cells and can simulate the liver metabolism process of in vivo drugs; the bottom chip is mainly connected with the left main channel and the right main channel through a collagen channel, and a brain glial cell layer and a brain microvascular endothelial cell layer are inoculated on a collagen interface to simulate the function of a blood brain barrier. The invention integrates the construction and characterization of an in vitro blood brain barrier model and the evaluation of the barrier permeability of the drug through the liver metabolic process, and can be used for the in vitro simulation of the blood brain barrier model and the evaluation application of the drug. The invention solves the problems of secondary inoculation and long time consumption of a cell co-culture model, the addition of the flow conditions better conforms to the real environment in vivo, the consumption of cells and reagents is obviously reduced, and a plurality of experiment related parameters can be simultaneously obtained at one time.
