abstract The invention provides application of an agent for inhibiting SMC1B gene expression in preparing a medicament for preventing and/or treating canceration of a hepatitis B patient. The invention also provides a kit for knocking out the SMC1B gene and a method for preparing an SMC1B gene knocked-out cell model. The inventor uses a specific sgRNA sequence to knock out SMC1B gene, so that the tumorigenicity of HBV can be reduced, especially the tumorigenicity of BVrtal 81T/sWl72 mutant strain is reduced very obviously, and the method has important theoretical and practical significance for preventing and treating clinical HBV related diseases.
