abstract The invention provides an asparagine endopeptidase small molecule inhibitor, which has a structural formula shown as follows: the invention also provides application of the small-molecule inhibitor of the asparagine endopeptidase in preparation of drugs for treating brain glioma. The small-molecule inhibitor of the asparaginyl endopeptidase can obviously inhibit the activity of the asparaginyl endopeptidase, and the activity of the asparaginyl endopeptidase is gradually reduced along with the increase of the concentration of the inhibitor. Meanwhile, in an intracranial glioma in-situ mouse model, the asparagine endopeptidase small-molecule inhibitor can obviously inhibit the tumor volume and obviously improve the survival rate and survival time of the mouse. Therefore, the small-molecule inhibitor of the asparaginyl endopeptidase can achieve the aim of treating tumor patients by effectively inhibiting the activity of the asparaginyl endopeptidase.
