A high-yield ansamitocin strain with enhanced polyketide synthase gene transcription level and a preparation method thereof belong to the technical field of biomedicine, and the ansamectin production is increased by increasing the expression level of the rate-limiting enzyme gene in the biosynthetic pathway. By using the erythromycin resistance gene promoter to enhance the expression level of the PKS gene ansa9 in the actinomycetes precious ATCC31280, the biosynthetic ability of ansamitocin is enhanced, and the production of ansamitocin is increased. The final ansamitocin fermentation yield of the engineering strain obtained in the present invention is about 125% higher than that of the original strain, and the laboratory shake flask level reaches 99 mg/L. |
