abstract The invention belongs to the technical field of sperm acrosome enzyme measurement, and discloses a human sperm acrosome enzyme kit, wherein gelatin mixed solution is prepared into sol matrix solution which is coated and fixed into a uniform basement membrane to display the activity of acrosome enzyme (Acroin); incubating sperms on an ultrathin gelatin basement membrane, centrifuging, washing with phosphate buffer solution to remove membrane inhibitory factor, depolymerizing sperm acrosomes, releasing and dissolving a gelatin matrix membrane to form a pore area surrounding the front part of the sperms; by light microscopic observation, light penetrates the aperture area, a bright halo is seen around the sperm head, an enzymatic reaction is observed under a dark purple blue background contrast after staining, and the size of the diameter of each bright ring is measured to estimate the intensity of the enzymatic activity of the acrosome of the individual sperm. The method is simple and low in cost, accurately calculates the positive reaction rate of acrosin, screens sperms with stronger activity for the test-tube infant technology, and improves the success rate.
