The method comprises the following steps: sterilizing the surface of a living sea horse, taking out an embryo or a liver under an aseptic condition, rinsing the embryo or the liver by using a rinsing culture solution, digesting the embryo or the liver by using trypsin, filtering to remove impurities, centrifuging, removing a supernatant, re-suspending cell precipitates by using a primary growth culture solution to prepare a single cell suspension, transferring the single cell suspension into a culture bottle added with the primary growth culture solution for culture, and replacing the primary growth culture solution by half of the culture solution every 2-3 days. The invention establishes a stable hippocampal primary cell culture system, and lays a foundation for research in the fields of hippocampal germplasm resource preservation, cell biology, immunology and the like. The primary hippocampus kelloggi cell culture system established by the invention is simple to operate, strong in repeatability and stable in cell growth, and can also be suitable for constructing other primary hippocampus kelloggi cell culture systems. |
