abstract | The invention discloses a histology method for distinguishing three muscle cells of carps, which comprises the following specific steps: freezing the treated sample by using isoprene and liquid nitrogen; freezing the sample at low temperature, slicing and drying at room temperature; putting the slices into constant-temperature pretreatment liquid for treatment, and then cleaning with cleaning liquid; adding cleaning liquid into the slices, pouring out after keeping the temperature constant, adding ATPase reaction liquid with the same temperature for reaction, and cleaning and soaking by using distilled water; sequentially adding CaCl to the slices 2 Solution, CoCl 2 Soaking the solution and ammonium sulfide solution, and cleaning and soaking the solution by using distilled water after the middle soaking is finished; and absorbing the redundant water of the section, covering a cover glass, sealing the section, and observing by using an optical microscope to distinguish the three muscle cells of the carp.
