abstract The invention relates to a construction method and application of a large fragment DNA library. The construction method of the large fragment DNA library provided by the invention is used for constructing the large fragment DNA library. The construction method of the large fragment DNA library comprises the steps of adding a 5 'end phosphorylation joint to a blunt-end DNA large fragment to obtain a joint-added DNA large fragment, cyclizing the joint-added DNA large fragment to obtain a mixture of a cyclized DNA large fragment and an unclycled DNA large fragment, and digesting linear DNA of the mixture to obtain a cyclized DNA large fragment, wherein the linear DNA digestion adopts exonuclease with a 5' end phosphorylation DNA specific digestion function.
