abstract | The invention discloses a preparation method of an ultra-thick tissue slice, which comprises the following steps: obtaining tissue material to be observed; preparing a tissue section of 1-8 mm of the tissue material and transparentizing the tissue section; and carrying out fluorescent staining on the transparent tissue section. Secondly, the method for reproducing the three-dimensional tissue morphological structure of the tissue slice comprises the following steps: preparing a transparent tissue slice with the thickness of 1-8 mm; acquiring image data of the tissue section by using a fluorescence microscope; recombining the image data to form three-dimensional model data; outputting a three-dimensional image based on the three-dimensional model data. The preparation method of the super-thick tissue slice and the method for representing the three-dimensional tissue morphological structure of the invention remove the fat of the tissue slice to be transparent so as to increase the depth of laser penetrating liver tissue and reduce light scattering, thereby realizing the depth imaging of the tissue, reflecting the cell structure of the tissue in a more real state and simultaneously improving the imaging efficiency and fidelity. |
