abstract The invention belongs to the technical field of livestock molecular marker screening, and particularly relates to a genetic marker associated with the lactation traits of Mediterranean buffaloes. The marker is obtained by screening buffalo ACAA1 gene, and the sequence of the marker is shown as SEQ ID NO:1, an A/T base mutation occurs at 776 base of the sequence, which results in ACAA1 gene polymorphism. The screening method comprises the following steps: extracting genome DNA from buffalo blood, designing primers, carrying out PCR amplification, cloning and sequencing PCR products, carrying out sequence comparison analysis, carrying out SNP detection, carrying out mass spectrum typing, and carrying out correlation analysis between markers and lactation traits. The SNP typing detection technology of the promoter region of the ACAA1 gene of the Mediterranean buffalo is disclosed, and the A/T mutation site is found to be obviously related to milk production traits such as 305d milk fat content, 306d milk protein content, 305 milk fat rate, 305 milk protein content and the like. The invention provides a new genetic marker resource for buffalo marker-assisted selection.
