abstract | The invention relates to a high-efficiency artificial transcription activator dCas9-TV and a coding gene and application thereof. According to the invention, multiple copies of VP64 and TAL transcription activation domains are connected to the carboxyl terminal of a Cas9 protein (dCas9) with inactivated nuclease, a series of novel artificial transcription activation factors are generated, and dCas9-TV with the best transcription activation activity is screened out. dCas9-TV efficiently activated transcription of endogenous genes in arabidopsis and rice when only one guide rna (grna) was used to target a specific gene promoter. When multiple grnas are used to target multiple target genes, dCas9-TV can simultaneously achieve transcriptional activation of multiple genes. In addition, dCas9-TV was demonstrated to have highly efficient targeted transcriptional activation activity in human cells as well. The in vitro assembled dCas9-TV/gRNA ribonucleoprotein complex can also be used for transcriptional activation of endogenous genes of arabidopsis thaliana and rice. By combining the excellent activities, the gene can be applied to aspects of genetic screening of genome, modification of biosynthetic pathway of metabolites, improvement of crops and the like. |
